Also to note I have tried accessing the Configuration utility both with the UAC enabled and Disabled and this has no affect. Please note that I also have Mass effect 1 and the same problem occurs with its configuration tool as well. The Mass Effect 1 game came from a retail box set. This makes it insanely hard to import my Mass effect 1 characters. If you use this tool to correct the location of the directory, and the program still isn't working properly, try selecting 'Refresh Tabs' under the 'Tools' menu. If the program still isn't working, it means some files are actually missing and need to be regenerated (see: Generate Missing Files), or another default location is incorrect (i.e., try using another tool in the 'Tools' menu).

After I had to wait to 5 PM EST for Mass Effect 2 to unlock, and then until 11 30 EST for Bioware to release enough unique product keys - THIS happened.
